No you do not , might spill a bit when you remove the trans but just plug the output shaft and you won't loose to much .

The transmission has nothing to do with the rear main seal, probably why no answers yet!

The rear main seal is on the engine, rear of the engine. Are you thinking of the seal at the end of the transmission?

+1, nothing to do with the tranny. Remove tranny and you'll see the engine rear main seal.

I would recommend you drain the trans fluid, it helps tremendously to tilt the engine upon removal of the transmission. The result is trans fluid leaking out of the tail shaft seal. If you prefer not to, a heavy duty latex glove, a little rtv, and a hose clamp works well.

Get the right tools and you won't have to tilt the engine to get the trans out. They make a cap for the trans tail shaft so you don't loose the fluid when you remove the driveshaft.

Good luck changing the rear main, they are fun lol
