Does the 2007 GT500's transmission benefit more with 410's as compared to 373's? I currently have 373's but had considered the swap. I do my own gear installation so other than the cost of the gears its not that expensive an upgrade.
Your 2007 has a 2.97 first vs a 2.66 first in the ‘13-14. Not exactly sure on your sixth but we have a 0.50 very tall final OD.
I think you’re a drag racer also so I try to gear the car for best trap speed. Not sure where you’ll be there, plus it’ll add more gear multiplication at launch, so you might be fine or better with a taller bias ply.
Hope that helps.
Regardless, these are heavy cars with 2 ODs in the trans, so the mechanical advantage of the 4.10s shine when the cars out of boost and at low speeds. Makes the car feel smaller and more responsive.

Here's some good information on transmission gearing :

When the 2007 Supercharged Shelby GT500 5.4L came along, it brought with it - you guessed it - another transmission! The Tremec TR-6060 6-speed is based on the T56, but improved from front to back. The aluminum case has an integral bell housing and the output yoke is a fixed design. Like its 05-10 GT sibling TR-3650, the 6060 uses a remote mount shifter. The 26-spline input shaft allows for much greater power handling. There have been three different ratio variations over the years. Gear ratios for the TR-6060 are:

Year 1st 2nd 3rd 4th 5th 6th Reverse
2007-2009 GT500 2.97 1.78 1.30 1.00 .80 .63 3.28
2010-2012 GT500 2.97 1.78 1.30 1.00 .74 .50 3.28
2013-2014 GT500 2.66 1.82 1.30 1.00 .77 .50 2.90

The TR-6060 can be found behind:
  • 2007-2012 Mustang Shelby GT500 5.4L 4V V8 Supercharged
  • 2013-2014 Mustang Shelby GT500 5.8L 4V V8 Supercharged

I got the information from here: What Transmission Is In My Mustang? - LMR.com
