My 2003 Cobra made 718/684 STD on a dynojet through a TH400 with 4k stall UCC spinning 325-50-15 M&H drag radials.

Power comes from a First gen FRPP 2.3 whipple. 2.90" upper 6lbs lower. Home ported & polished inlet.
JLT big air intake.
.020" over stock block. Stock SVT rods & crank.
flat top diamond pistons with reliefs. 10.5:1 static compression.
ARP head & main studs.
Cometic head gaskets.
Ported heads, FR500 cams setup with a lot of split.
JBA mid length headers to 3" X pipe, 3" bullets turned down at the axle.
ID1000s maxed at 96% DC despite 60psi deltaP.
2 Jayracing Pro 044 pumps & wire kits.

25lbs up to about 5300 rpm's and down to 20lbs by 6500 rpm's.

Belt slip after 5300 rpm's. Should be making quite a bit more power when I can get the belt to hold the boost all the way up to 7k rpm's.

Is it safe to assume tuning a Supercharged Mustang to run E85 increases an ave of 100hp?

too many variables to come up with a "average" number IMO

typically when you run E85 you bump the boost and timing so not all things are equal.

Over pump gas a 100rwhp gain is possible with an aftermarket power adder. In fact, E85 will allow a combination that can't reliably even be run with pump gas (flat top pistons, >10:1 static compression, and >20psi boost with >15° timing advance)

For an Eaton I don't think you'd see a 100rwhp gain but it could be possible.

Compared to race gas with a >100 octane, there won't be much gain with E85, but you can get away with higher IAT2 due to the impressive cooling nature of E85, as well as the 30% increase in volume.

However, there is still a threshold that you don't want to go over. Hot spots can cause pre-ignition without audible detonation in an E85 compression event. You'll just go from running hard to total loss of compression due to torched piston and/or head. It's unclear to me if a J&S knock sensor/guard can detect pre-ignition in an E85 compression event and I can't afford to fund the research to find out...

I saw 25rwhp on my stock motor / eaton combo through an auto from VP109 to E85. Same mods, same boost...E85 took 1 more degree of timing though.

Anybody notice that e85 went up over a dollar/Gal last week? I checked 3 stations in 3 different countys and they are all up....WTF?
